| RayBio© Enzyme Immunoassay (EIA) kits are
competitive, single-antibody quantitative immunoassays that
are particularly useful for detecting small peptide hormones that are more difficult to detect using traditional sandwich ELISA assays. Signal intensities measured in the EIA assay are subject to competitive binding to a common target-specific capture antibody by biotinylated peptide standards and unlabeled peptide/protein in standards or samples. |

| RayBio© EIA
microplates are pre-coated with a 2°
antibody that recognizes the target-specific capture antibody
used in the assay. After blocking, the capture antibody is
incubated in each well, followed by the addition of either an
unknown sample or unlabeled peptide standard of known
concentration. Additionally, each sample and standard is
spiked with equal amounts of a biotinylated peptide standard.
Simultaneous incubation of both the biotinylated peptide
standard and unlabeled target proteins/peptides in the samples
or unlabeled peptide standards (added in various
concentrations to for the calibration curve) results in
competition for binding to the available capture antibodies in
each well. After this, streptavidin-conjugated horseradish
peroxidase (HRP-streptavidin) is added to each well, which
binds to the biotin-labeled peptide, and detection is
accomplished via colorimetry with TMB, a chromogenic
substrate for HRP commonly used in ELISA assays.
Since the colorimetric signals result solely from binding
of HRP-Streptavidin to biotinylated peptides, signal
intensities measured in the assay are directly proportional to
the amount of biotinylated peptide bound to the capture
antibody and are inversely proportional to the amount of
unlabeled target protein/peptide in standards or samples. A
calibration curve can be established then using signal
intensities resulting from competition by known concentrations
of unlabeled peptide standards, and the concentration of
target protein/peptide in each sample can be calculated based
upon comparison of colorimetric signal intensities to those of
the known standards.. |
