Quantibody
array is an array-based multiplex sandwich ELISA system for simultaneous
quantitative measurement the concentration of multiple cytokines in serum,
plasma, CSF, cell lysate, conditioned media and other body fluid. It combines
the advantages of the high detection specificity/ sensitivity of ELISA and the
high throughput of the arrays. Like a traditional sandwich-based ELISA, it uses
a pair of cytokine-specific antibodies for detection. A capture antibody is
first bound to the glass surface. After incubation with the sample, the target
cytokine is trapped on the solid surface. A second biotin-labeled detection
antibody is then added, which can recognize a different isotope of the target
cytokine. The cytokine-antibody-biotin complex can then be visualized through
the addition of the streptavidin-labeled Fluorescent dye using a laser scanner.
Unlike the traditional ELISA, Quantibody products use array format. By arraying
multiple cytokine specific capture antibodies onto a glass support, multiplex
detection of cytokines in one experiment is made possible.
In detail, one
standard glass slide is spotted with 16 wells of identical
cytokine antibody arrays. Each antibody, together with the
positive and negative control is arrayed in quadruplicate.
The slide comes with a holder that includes a 16-well
removable gasket that allows for the processing of up to
16 samples on one slide. Four slide holders can be nested
into a tray, which matches a standard microplate and
allows for automated robotic high-throughput process of 64
arrays simultaneously.
For cytokine
quantification, the array specific cytokine standards,
whose concentration has been predetermined, are provided
to generate a five-point standard curve of each cytokine.
In a real experiment, standard cytokines and samples will
be assayed in each array simultaneously through a sandwich
ELISA procedure. By comparing signals from unknown samples
to the standard curve, the unknown cytokine concentration
in the samples will be determined.

Less
sample, more data:
Only
50 l sample is needed for quantification of up to 20
cytokines in quadruplicate. This is 80 times more
efficient than traditional ELISA. The advantage of
Quantibody array is even precious when sample supply is
limited.
Less
time
The
processing time for one slide is around 4 hours. Because
of the high-throughput capability of the array, hundreds
of samples can be processed by one technician in a single
day.
Much
lower cost
In
order to get triplicate data results, the average cost per
cytokine per sample for traditional ELISA is normally over
$13 in the 96-well plate format. However, in Quantibody
Arrays, even the quadruplicate data results will only cost
less than $3 per cytokine per sample for a 10-cytokine
array. This is much affordable and cost effective.
ELISA
detection sensitivity
ELISA
is well-known for its sensitivity and specificity in
cytokine quantification. Our Quantibody products use
sandwich-ELISA based methology and have been tested to
have similar detection sensitivity as traditional ELISA.
Reproducibility
Quantibody
products use photobleaching resistant Alexa dyes for
detection, which greatly increases the system
reproducibility and dynamic detection sensitivity. The
slides can be rescanned for multiple purposes and stored
in dark for months without loss of sensitivity.
Flexibility
Our
available kits include either 10 or 20 defined cytokines
in the detection panel. However, our custom arrays enable
our users to choose not only the cytokine kinds but also
the number of cytokines to be included in the array from
our several hundred of cytokine pools. In addition, users
can choose Alexa 647 dye (red, cy5 channel) for detection
instead of Fluorescent dye (green, cy3 channel).
Quantitative
analysis software available
Quantibody
array-specific Q Analyzer is available for data
computation. It is an Excel-based program, which is easy
to operate and requires no professional training. Simply
copy and paste the raw data into the assigned area. The
program will automatically remove the outlier spots and
output standard curves and sample cytokine concentrations.

What is included in the product?
Our
Quantibody product is provided as a kit, which includes
all the necessary reagents for performing the experiments.

What equipment do I need for imaging?
A
standard glass slide laser scanner with cy3- or
cy5-compatible wavelength is needed for the signal
detection. User can choose Fluorescent dye (green,
cy3 channel, 555nm excitation, 565nm emission) signal detection.

What kind of samples can be detected with
Quantibody kits? And how much sample do I need for the
experiment?
Quantibody
arrays can be used in cell culture supernatant, serum,
plasma, CSF, tissue lysate, and other body fluid.
Typically, 50-100 μl samples are needed for one
array.
We
recommend the following parameters for your samples:
50-100 μl of cell culture supernatant, 50-100 μl
of original or diluted serum or plasma; or 20-200 μg
of protein for cell lysates and tissue lysates. In order
to minimize the matrix effects and to lower the background
of the assay, we recommended that the samples at least
diluted 2 fold with Sample Diluent. Dilute the lysate at
least 5 fold with Sample Diluent to make a total volume of
50 to 100 μl. For those samples in which you expect a
high response for one cytokine and a low response for
another, we recommend testing each sample in step
dilutions. For example, when testing cell culture
supernatant for unknown cytokine concentrations; a 1:2, a
1:10, and a 1:50 dilution are typically used.
4. What cytokine
standards are used in the Quantibody Array?
Laboratories
throughout the world use different bioassays and
immunoassays to determine cytokine concentrations. As a
result, the reported cytokine concentrations in the same
sample may vary when using different ELISAs developed by
different labs. For this reason, the availability of
international standard preparations of cytokines is
essential to allow definitive analysis and comparison of
results. Quantibody Arrays use either our internal
cytokine standards or the International Biological
Standards from the National Institute for Biological
Standards and Control (NIBSC), the gold cytokine
standards.

Is there any cross-reactivity between the cytokines?
For the available kits, no.
. We have tested
all of the available antibodies with their specific
purified antigens for the cross-reactivity. The
antibody pairs used in the kit have been tested to
recognize their specific cytokines. There
may be some cross-reactivity between antibodies included
in two different kits, but analysis of samples
containing only a single recombinant cytokine found no
cross-reactivity with other antibodies in the same array. For
the custom arrays, we will suggest what cytokines can be
included in the same array to ensure the proper level of
specificity and sensitivity.

Can I save the remaining cytokine standard for next
time use?
The
cytokine standards in the kit are provided in the
lyophilized mixture form to ensure the stability. For the
quality purpose, the cytokine standards once reconstituted
should be used in a short period of time (2 3 days) and
cant be refrozen because they may lose activity during
the freeze-thaw cycles. For those in need, we suggest only
save the highest concentrated vial, and re-do the serial
dilution for a second time usage.

Do I need special software to quantify my data?
Yes
and no. After imaging by a laser scanner and data
extraction with any of the microarray analysis software (GenePix,
ScanArray Express, ArrayVision, or MicroVigene), cytokine
quantification can be done with virtually any ELISA
analysis software. However, our array-specific Q Analyzer
software is available to facilitate the data analysis.
Instead of tedious calculation, users can now quickly
figure out the unknown sample concentration through a
simple copy and paste process. The program can
automatically remove the outlier spots for greater
accuracy.
8.
I would like to try Quantibody products but I
dont have access to a laser scanner. What can I do?
Two
options. You may order the Quantibody Arrays and do all
the experiments yourselves, and then ship back the slides
to RayBiotech Inc. For a certain fee, RayBiotech will do
the imaging and all the data analysis for you. Or for
quality purpose, we offer a custom processing service.
Just sending in your samples, and we will process the
arrays and send back data within 5-10 business days.
Service includes: Sample processing with our experienced
technicians; Slide imaging & data extraction with our
laser scanner and data extraction software; Cytokine
quantification with Q Analyzer software; Return results
include image and the analysis results in Excel format.
