Cell-based assays use live cells to offer a biologically relevant model one can use to track dynamic variables or functional or biochemical effects of cells and their behavior. Human peripheral blood mononuclear cells (PBMCs) are peripheral blood cells having a single round nucleus including lymphocytes, monocytes, natural killer cells (NK cells) or dendritic cells (DCs). PBMCs are isolated from blood based on its gravity different from other blood cells, e.g. red blood cells and granulocytes. Because PBMCs contain the most important immune cells responding to foreign challenges, especially lymphocytes, they are widely used in immune research in vitro. Either PBMCs from normal donors or from diseased individuals are used to investigate phenotypes of different immune cells at physiological or pathological conditions, the data of which can be used for diagnosis of certain diseases including infectious disease, autoimmune disease, etc. PBMCs are also used to enrich a specific type of immune cells e.g. T cells, B cells or monocytes, so that scientists can observe their functions at different conditions. In vitro stimulation of PBMCs with various of agents, which mimics a immune response, is widely used a model to investigate the mechanism of T cell or B cell activation, and to find ways to enhance “good” immune response, e.g. anti-viral infection immune response, and inhibit “bad immune response, e.g. autoimmune disease.
