Applications:
ELISA (recommended work dilution= 1: 1,000-2,000), Western Blotting (recommended work dilution= 1~5 µg/ml), Dot Blotting(recommended work dilution= 1~5 µg/ml), (see image)The Purity and binding activity of recombinant antibody, anti-O157:H7 by Dot-blotting assay. A. O157 scFv binding activity. Directly spotting O157 antigen into membrane in Row I and II with two dilution (100 and 10 ng of each spot 2-3 ) plus positive system control in spotting blot 1 and 5 as well as BSA control in Spot 4, respectively, then probe the membrane by using O157:H7-specific recombinant antibody after blocking and following anti-Myc-tag and HRP-conjugated detection antibody. B. Checking purity of purified O157 scFv. Directly probed blocking blot with anti-Myctag and following HRP-conjugated detection antibodies.. Anti-Myc Tag Antibody Applications: Immunoassay (ELISA, Western blotting)(recommended work dilution= 1:2,000-1:5,000)

Description

Escherichia coli O157:H7 is an enterohemorrhagic strain of the bacterium Escherichia coli and a cause of foodborne illness. Infection often leads to bloody diarrhea, and occasionally to kidney failure, especially in young children and elderly people. Most illness has been associated with eating undercooked, contaminated ground beef, drinking unpasteurized milk, swimming in or drinking contaminated water, and eating contaminated vegetables.
A human recombinant antibody to O157:H7 was expressed in E. coli and purified using NiNTA affinity chromatography. The antibody is tagged with a myc-tag (EQKLISEEDL) and a his-tag (HHHHHH) at the C-terminus of the antibody heavy chain.

Storage / Stability

The antibody is stable for at least 1 year from the date of receipt when stored at -20°C to -70°C. Reconstituted antibody can also be aliquotted and stored at 4°C for 1 month or at -20°C to -70°C in a manual defrost freezer for many months without detectable loss activity. Please avoid freeze-thaw cycles.

Specificity

It specifically detects protein derived from testing bacterial sample at various concentrations. See image Below.
