Preparation:
Immunogen was synthetic peptide derived from Human/Mouse Angiotensin II. This antibody was produced from a rabbit immunized with the immunogen. The IgG fraction was purified from rabbit serum by ammonium sulphate precipitation followed by Protein A/G affinity chromatography.
Reconstitution:
A separate vial of dilution buffer is provided for reconstitution. The antibody is supplied lyophilized, originally containing PBS, without preservative stabilizers (e.g. sodium azide). The final amount is indicated on the shipping vial.
Angiotensin II (Ang II) is the key peptide hormone in the renin-angiotensin-aldosterone system (RAAS). Its ability to regulate levels of circulating aldosterone relies on actions on adrenal glomerulosa cells. Many of the Ang II effects on glomerulosa cells involve a precisely coordinated regulation of signaling cascades and gene expression. Ang II interacts with its G protein-coupled receptor in zona glomerulosa adrenal cells and triggers a cascade of intracellular signals that regulates steroidogenesis and proliferation. It also promotes vascular smooth muscle cell (VSMC) growth, stimulates Ca(2+)- calmodulin (CaM)-dependent kinase II (CaMKII), and activates cytosolic Ca(2+)-dependent phospholipase A2 (cPLA2), which releases arachidonic acid (AA). ANG II also generates H2O2 and activates Akt, which have been implicated in ANG II actions in VSMC.
The antibody is stable for at least 1 year from the date of receipt when stored at -20°C to -70°C. Reconstituted antibody can also be aliquotted and stored at 4°C for 1 month or at -20°C to -70°C in a manual defrost freezer for many months without detectable loss activity. Please avoid freeze-thaw cycles.
The rabbit anti-human/mouse Angiotensin II antibody detects specifically target at various concentrations in ELISA and EIA assays. Cross reactivity with rat is expected from sequence similarity
