Preparation:
Immunogen was synthetic peptide derived from Human Ghrelin precursor. This antibody was produced from a rabbit immunized with the immunogen. The IgG fraction was purified from rabbit serum by ammonium sulphate precipitation followed by Protein A/G affinity chromatography.
Reconstitution:
Supplied as 200 µg of purified and lyophilized antibody. The antibody is stable for at least one year from the data of receipt when stored at -20°C to -70°C. Reconstituted antibody (recommended with H20 1 mg/ml) can also be aliquoted and stored frozen at -20°C to -70°C in a manual defrost freezer for months without detectable loss activity. Upon reconstitution, the antibody can be stored at 4°C for 1 month. If 1 ml of H20 is used, the antibody concentration will be 200 µg/ml. Please avoid freeze-thaw cycles.
Description
Ghrelin is synthesized as a preprohormone, and then proteolytically processed to yield a 28-amino acid peptide. It is a recently isolated brain-gut peptide that has growth hormone-releasing and appetiteinducing activities. Several recent studies have suggested that ghrelin plays a major role in the pathophysiology of drug-seeking behavior and anxiety. The rat and human mature Ghrelin (28-aa) are produced from 117 amino acids precursor peptides that have a signal peptide followed by the ghrelin sequence. At the C-terminus of the ghrelin sequence, processing occurs at an uncommon Pro-Arg recognition site. In rat stomach, two isoforms of mRNA for pro-ghrelin are produced from the gene by an alternative splicing. One mRNA encodes the ghrelin precursor, and another encodes a des-Gln14- ghrelin precursor. O-octanoylation or O-decanoylation is essential for ghrelin activity. The Odecanoylated forms Ghrelin-27-C10 and Ghrelin-28-C10 differ in the length of the carbon backbone of the carboxylic acid bound to Ser-26.
Specificity
The rabbit anti-human Ghrelin antibody detects specifically target derived from related-cell lysates protein at various concentrations. Cross reactivity with mouse and rat are expected from sequence similarity.
Storage
Store at -20°C, if not intended for use within a month, Keep it at 4°C and minimize freezing and thawing when use.
