Preparation:
Immunogen was synthetic peptide derived from Human Hepcidin. This antibody was produced from a rabbit immunized with the immunogen. The IgG fraction will be purified from rabbit serum by ammonium sulphate precipitation followed by Protein A/G affinity chromatography.
Reconstitution:
Supplied as lyophilized purified antibody. The antibody can be reconstituted in sterile PBS, be stored at 4°C for 1 month after resuspension. Please avoid freeze-thaw cycles.
Hepcidin, also called as LEAP-1 is cysteine-rich peptide containing four disulfide bonds. Its active form, 25-residue peptide was identified in human blood. It was predominantly detected in the liver, and, to a much lower extent, in the heart. Gram-positive Bacillus megaterium, Bacillus subtilis, Micrococcus luteus, Staphylococcus carnosus, and Gram-negative Neisseria cinerea as well as the yeast Saccharomyces cerevisiae dose-dependently exhibited sensitivity upon treatment with synthetic Hepcidin. It is also the central regulator of systemic iron homeostasis. Dysregulation of hepcidin production results in a variety of iron disorders. Hepcidin deficiency is the cause of iron overload in hereditary hemochromatosis, iron-loading anemias, and hepatitis C. Hepcidin excess is associated with anemia of inflammation, chronic kidney disease and iron-refractory iron deficiency anemia.
The antibody detects specifically synthetic immunogen. Cross reactivity with mouse and rat Hepcidin are not tested yet.
Store at -20°C, if not intended for use within a month, Keep it at 4°C and minimize freezing and thawing when use.
