Applications:
Direct and indirect ELISA: antibody detection limit dilution at least 1:1,000-2,000. Western Blot: Reacted with the approx 48-kDa Vaspin His tagged protein. Suggested blocking and dilution buffer is PBST with 0.05% Tween 20 with 5% skim milk. Suggested incubation time is 1 hour at room temperature. Recommended for use at dilution 1:500-1,000.
Preparation:
Immunogen was synthetic peptide derived from Human Vaspin. This antibody was produced from a rabbit immunized with the immunogen. The IgG fraction was purified from goat serum by ammonium sulphate precipitation followed by Protein A/G affinity chromatography.
Reconstitution:
A separate vial of dilution buffer is provided for reconstitution. The antibody is supplied lyophilized, originally containing PBS, without preservative stabilizers (e.g. sodium azide). The final amount is indicated on the shipping vial.

Visceral adipose tissue-derived serpin (vaspin) was identified as a member of serine protease inhibitor family, which was expressed in visceral adipose tissue of OLETF rats at the age when obesity and insulin plasma concentrations reach a peak. Vaspin expression was shown to decrease with worsening of diabetes and body weight loss, whereas vaspin serum levels could be normalized by insulin or pioglitazone treatment. Administration of vaspin to obese mice improved glucose tolerance, insulin sensitivity, and altered gene expression of candidate genes for insulin resistance. Induction of human vaspin mRNA expression in adipose tissue is regulated in a fat depot-specific manner and could be associated with parameters of obesity, insulin resistance, and glucose metabolism.
The antibody is stable for at least 1 year from the date of receipt when stored at -20°C to -70°C. Reconstituted antibody can also be aliquotted and stored at 4°C for 1 month or at -20°C to -70°C in a manual defrost freezer for many months without detectable loss activity. Please avoid freeze-thaw cycles.
The rabbit anti-human Vaspin antibody detects specifically target derived from human recombinant protein and Hela cell lysate at various concentrations. Cross reactivity with mouse and rat are expected from sequence similarity.
