This kit utilizes a DNA spin column and a novel solution system, suitable for the rapid and simple extraction of genomic DNA from plant samples containing polyphenols, polysaccharides, and enzyme inhibitors. It can purify DNA from one or more 100 mg fresh or 20 mg dried plant samples in 30 minutes. The extraction process does not require toxic organic solvents such as phenol or chloroform, nor time-consuming isopropanol or ethanol precipitation. It efficiently removes impurities such as polysaccharides, polyphenols, and enzyme inhibitors. The purified DNA is suitable for PCR, enzyme digestion, and hybridization experiments.
Fresh or dried plant tissue (cells) are ground and lysed; proteins, polysaccharides, and cell debris are precipitated and removed. Genomic DNA is then selectively adsorbed onto the silica membrane in the spin column under high salt conditions. A series of rapid wash-centrifugation steps further remove polysaccharides, polyphenols, cell metabolites, and proteins. Finally, a low-salt elution buffer elutes the purified genomic DNA from the silica membrane.
