Human macrophages were generated by culturing PBMC-derived monocytes with 50 ng/ml MCSF for 6 days. The macrophages were polarised to M2c in the presence of 50 ng/mL IL-10 and TGF-β for an additional 2 days. The cells were collected and incubated with 100 nM of the anti-human CD32 antibody for 30 min on ice, followed by 30 min of a-mouse AF647 secondary antibody. The expression of CD32 was analyzed by flow cytometry.
