We ran 3-D cell culture supernatants were run with the kit for the first time as feasibility. We saw no results across any kidney cell culture samples regardless of time point. Extensive optimization with the dilution may be required.

Measured human plasma samples at 4-fold dilution and human urine at 4-fold and 20-fold dilution. We were able to detect podocalyxin in both biofluids at all dilutions. Observed a matrix effect for plasma requiring samples to be diluted further.

I used this product to measure podocalyxin concentrations in human urine samples. The manual was easy follow, and includes every small step (sometimes more than once to help avoid errors).The suggested podocalyxin concentrations for constructing the standard curve worked pretty well, except the highest concentration (which saturated our reader).
