Introduction
Aldolase is a crucial enzyme in glycolysis and gluconeogenesis for energy production and glucose synthesis. It exists as three isoenzymes: Aldolase A (skeletal muscle and red blood cells; an indicator for late-stage muscular dystrophy, cancer, and myocardial infarction), Aldolase B (liver, kidney, and small intestine; associated with cirrhosis, hepatitis, and hereditary fructose intolerance), and Aldolase C (brain; associated with encephalitis, cerebral infarction, and some cancers). Monitoring aldolase levels is important in diagnostics and disease monitoring.
Assay Principle
Based on a coupled enzyme reaction, fructose-1,6-bisphosphate is catalyzed by aldolase to a colored product quantified at 450 nm. The change in absorbance is directly proportional to aldolase activity in the sample.
