• Pipette tips (barrier tips recommended) and microcentrifuge tubes
• PBS
• Deionized (DI) Water
• Microcentrifuge
• Vortex mixer
• Precision pipettes to deliver 2 µL to 1 mL volumes
• Tissue homogenizer if using tissue samples (Dounce tissue grinder recommended)
• Refrigerated centrifuge with swing out rotor and refrigerated microcentrifuge
• Optional: PMSF (Phenylmethyl sulfonyl fluoride) or similar serine protease inhibitor

1. Prepare cell or tissue samples as instructed.
2. Prepare reagents as instructed.
3. Sequentially isolate cytoplasmic, nuclear, and histone fractions.
4. Proceed through the workflow; extend histone extraction overnight if needed.

The kit can be stored for up to 12 months in 4°C except for the Phosphatase Inhibitor Cocktail (store at -20°C or -80°C). After Protease Inhibitor Cocktail has been reconstituted, store at -20°C for up to 5 days.
