E. coli LPS ELISA kit Standard Curve
The LPS ELISA kit was used for a ligand binding assay to assess interaction between lipids and LPS. The LPS used was from Sigma and hence, a standard curve was designed with the LPS ELISA to check detection of the coated antibody to the LPS used. The LPS ELISA did detect the Sigma LPS and the standard LPS provided with the kit in a similar manner and standard curve with both standards was comparable. Sample: Mixture of Lipid + LPS at different concentrations Standard ELISA concentrations: 2ng/mL, 4ng/mL, 5ng/mL, 6ng/mL 8ng/mL, 10ng/mL and 12ng/mL Experimental conditions: Same as the instruction provided in the kit. The LPS ELISA instructions were clear, easy to follow and can be used for ligand binding assays. However, the sensitivity of the ELISA falls at 2ng/mL and hence, should be used for detections at concentrations above 3ng/mL. As per the instructions manual, the substrate needs to be incubated for 30 min. However, an incubation between 15-20 min was sufficient for robust color determination.
