Mariculture Keyhole Limpet Hemocyanin (mcKLH) is a highly stable and effective carrier protein, commonly used to generate antibodies against haptens and peptides. Its large molecular size and complex structure provide numerous reactive sites for efficient conjugation with peptides and other antigens using amine- or carboxyl-reactive crosslinkers. As a result, mcKLH is widely recognized as a standard carrier protein for antibody production against haptens and peptides.

Hapten-mcKLH Conjugation Kit provides an optimized method for preparing hapten-carrier conjugates for immunization and antibody production. This kit employs a carboxyl-reactive carbodiimide crosslinker, which forms stable bonds between exposed carboxyl and amino groups on peptides and proteins. mcKLH is pre-formulated in buffers compatible with carbodiimide chemistry and the use of spin desalting columns ensures exceptional protein recovery through a simple centrifugation step.

• Hapten such as DNA or RNA oligonucleotide, small molecule, peptide or small protein containing a primary amino group
• Centrifuge
• Sterile nuclease-free pipette tips (barrier tips recommended) and 1.5 mL microcentrifuge tubes
• 15 mL tubes
• Nuclease-free water
• PBS (optional)
• DMSO (optional)

1. Resuspend one vial of mcKLH in nuclease-free water.
2. Dissolve hapten (up to 2 mg) in Conjugation Buffer.
3. Add the hapten solution to the mcKLH solution to make Hapten-mcKLH solution.
4. Resuspend one vial of EDC in the Hapten-mcKLH solution and mix gently.
5. Prepare the Spin Desalting Column by removing the storage solution and washing with Purification Buffer.
6. Apply the Hapten-mcKLH-EDC solution to the column and centrifuge to collect the conjugate.
7. Quantify the Hapten-mcKLH conjugate by BCA protein assay.
8. The Hapten-mcKLH conjugate can be used for immunization or stored for later use.

The entire kit may be stored at 2 - 8°C. Do not freeze. The kit can be stored for up to 12 months from the date of shipment. Minimize light exposure.
