The mitochondrial genome (mtDNA) is a circular, intron-free, double-stranded, haploid DNA strand 16.5 kb in length that encodes 37 genes. mtDNA copy number and damaged are biomarkers for mitochondrial dysfunction associated with overall mortality and a number of age-related diseases, including cardiovascular disease, chronic kidney disease, and cancer.
RayBio® Human Hela cell Mitochondrial DNA (MTH-HDNA) is isolated from the Hela cell line using the RayBio® Mitochondrial DNA Isolation Kit. The quality and purity of mtDNA are validated by gel electrophoresis, ratio of mtDNA and genomic DNA, and mtDNA damage quantification kit. The quantity of mtDNA was measured by the RayBio® dsDNA Quantification Assay Kit. The mtDNA is provided ready to use for a variety of mitochondrial analyses including PCR, SNP analysis, copy number, damage and function assay.

The high-quality mtDNA is free of contaminants of nuclear DNA and RNAs. Purified mtDNA has high quality metrics, including A260/A280 = 1.8~2.0 and A260/A230 > 2.0. It typically has a single peak size of 16.6 kb, making this kit an excellent choice upstream of long-read sequencing platforms.
The mtDNA can be stored at -20°C for a period of 12 months upon arrival. After opening, the reagents are valid for at least 6 months if stored at -20°C. For longer stability, we recommend to aliquot, store mtDNA at -80°C and avoid repeated freeze/thaw cycles.
