Introduction
Protein lactylation plays an unusually prominent role in cell signaling, development, and growth. The RayBio Human Protein Lactylation Antibody Array 2 is a very rapid, convenient, and sensitive assay that can simultaneously detect multiple lactylation events on proteins. It can also be used to monitor the activation or function of important biological pathways.
RayBiotech is committed to developing a series of lactylation antibody arrays. The RayBio Human Protein Lactylation Antibody Array 2 is specifically designed for the simultaneous identification of the relative levels of lysine lactylation in 493 different human proteins in cell lysate. By monitoring changes in protein lactylation in your experimental model system, you can verify pathway activation in your cell lines without the need for extensive time and effort required for immunoprecipitation and/or Western Blot analysis.
To use the RayBio Human Protein Lactylation Antibody Array 2, treated or untreated cell lysate is added to the antibody array glass slide wells. The antibody array slide wells are then washed, and a biotinylated anti-Lactyllysine antibody is used to detect the lactylated lysine residues on target proteins. After incubation with a fluorescent dye-conjugated streptavidin (Cy3 equivalent), the slides can be imaged using a laser scanner, such as the Axon GenePix, in the Cy3 channel.

Assay Procedure Summary
- Dry the glass slide
- Add Blocking Buffer to each well and incubate.
- Add the sample to each well and incubate.
- Wash the wells.
- Add Biotinylated Detection Antibody solution to each well and incubate.
- Add Fluorescent Dye-Conjugated Streptavidin solution to each well and incubate.
- Scan with a gene microarray laser scanner
- Perform densitometry and analysis
