Telomere length (TL) and mitochondrial DNA copy number (mtDNA-CN) are widely recognized molecular biomarkers of biological aging and all-cause mortality. Oxidative stress and DNA damage drive telomere attrition and mitochondrial dysfunction in parallel, linking both markers to cumulative cellular stress. Short telomeres suppress mitochondrial biogenesis and are strongly associated with cardiovascular disease, chronic kidney disease, cancer, and other age-related disorders. In contrast, the combined profile of short TL with elevated mtDNA-CN is emerging as a signature of accumulated damage accompanied by compensatory mitochondrial proliferation. Reliable, simultaneous measurement of TL and mtDNA-CN is therefore critical for aging research, preclinical drug development, and translational studies.
The RayBio® Human Telomere Length and Mitochondrial DNA Copy Number Dual Quantification Kit is a TaqMan™ probe-based duplex qPCR system with highly specific primer–probe sets to selectively amplify telomeric repeats and the mitochondrial ND1 gene while excluding nuclear genomic loci. A single-copy reference (SCR) gene enables robust internal normalization for either absolute or relative quantification. A reference DNA standard with certified TL and mtDNA-CN values is included for assay validation and consistency.
The single-tube duplex reaction provides linear quantification across at least five orders of magnitude and is compatible with DNA isolated from cultured cells, tissues, whole blood, saliva, urine, lavage fluids, cerebrospinal fluid (CSF), or scraped cells. Both TL and mtDNA-CN measurements can be generated in under two hours, streamlining high-throughput and time-sensitive workflows.

The kit can be stored at -20°C for a period of 12 months prior to opening. After opening, the reagents are valid for at least 6 months if stored at -20°C. Avoid repeated freeze-thaw cycles.
