Enrichment of mitochondrial, cytosolic, and nuclear fractions from cultured HeLa and BHK cells.
Cultured Human Ovarian Cancer cell OV433 samples was prepared as instructed in the manual. Prepared all reagents and keep buffers on ice during the procedure. Divided the prepared sample into a whole cell extract control and a fractionation sample. Sequentially isolated the cytosolic, mitochondrial, and nuclear fractions from the fractionation sample. Stored the enriched fractions at -80°C until use. We got good protein detection for the Lysis Buffer II and Lysis Buffer III Lysates. And we like Protease Inhibitor Cocktail. But, we have problem with Lysis Buffer I Lysate. We couldn't detect any b-Actin protein band from the suggested 500ul lysate volume even after we optimized the cell counts to 1X10^7 and 6X10^6 and prolonged lysis time and passing through the pipette many times (totally 50 times). We checked the lysate under the microscope and saw many rounded cells even after repeated lysis steps. Is buffer I too weak to lysis the cells?
