The RayBio® Mouse IL-6 ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse Interleukin 6 in serum, plasma and cell culture supernatants. This assay employs an antibody specific for mouse coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-6 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse IL-6 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-6 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm
Interleukin 6 is a pleiotropic cytokine that regulates cell growth and differentiation of various tissues, and plays an important role in the immune response and acute phase reactions. It acts as both a pro-inflammatory and anti-inflammatory cytokine. It is secreted by T cells and macrophages to stimulate an immune response, e.g., during infection and after trauma, especially burns or other tissue damage leading to inflammation. IL-6's role as an anti-inflammatory cytokine is mediated through its inhibitory effects on TNF-α and IL-1, and activation of IL-1Rα and IL-10.

Recently used the IL-6 ELISA kit for quantifying interleukin-6 (IL-6) levels in cachexia mouse plasma samples. The kit came with all necessary reagents, including pre-coated 96-well plates, standards, detection antibodies, and substrates, which minimized preparation time and potential errors. The IL-6 ELISA kit protocol was straightforward, and all the assay incubations were at RT. The dilutions (2-fold) worked good as suggested in the protocol. Will order this kit for our experiments further.

We've been looking for an ELISA kit that detects IL-6 in tissue lysates for ages. We used this kit with mouse brain tissue homogenized in a lysis buffer containing 0.25% SDS and diluted the samples 1:5 as recommended by the manufacturer protocol and got beautiful results.

I have used this Elisa kit, its quite affordable considering the amount of time saved and data generated from it. I have even compared IL-6 ELISA data to IL-6 luminex data (from the same samples) and they are relatively similar.

I used this kit with LPS-inflammed RAW264.7 cells supernatants. It was very sensitive! The instructions were clear and detailed and the kit well assembled. I also liked that you can use just half a plate and keep the other strips for another time. I would strongly recommend.

The kit was packaged in an organized and clear manner, and it was easy to walk through each step of the procedure. I used the mouse IL-6, VEGF, and TNF-alpha kits for plasma as well as homogenized muscle samples. Each worked well, and I would be happy to use them again.
