The RayBio^® YTE-Fc Mutations ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of free drug antibodies containing YTE mutations in the Fc region (M252Y, S254T, and T256E triple substitution) in human samples, cell culture supernatants, and purified biological products.

Note	This ELISA uses an antibody pair and standard that have been validated for multiplex array products, but not for the 96-well plate ELISA format. Thus, this ELISA requires a 4 - 7 week development phase, which RayBiotech begins after receiving an order for the kit. After development, the kit will have passed RayBiotech's ELISA quality control tests, and will thereafter have a typical 1 - 2 day lead time for orders placed.

Assay Procedure Summary

Prepare all reagents, samples and standards as instructed in the manual.
Add 100 µL standard or sample to each well. Incubate 2.5 hours at room temperature.
Add 100 µL prepared biotin antibody to each well. Incubate 1 hour at room temperature.
Add 100 µL prepared Streptavidin solution to each well. Incubate 45 minutes at room temperature.
Add 100 µL TMB One-Step Substrate Reagent to each well. Incubate 30 minutes at room temperature.
Add 50 µL Stop Solution to each well. Read at 450 nm immediately

Storage/Stability

The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.
