I am very pleased with this ELISA kit. I wanted to measure incretins in unfasted mouse plasma (i.e., mice were free-living, and not specifically fasted or stimulated with glucose, food, etc. prior to blood collection). The plate kit comes with plenty of extra reagents to allow for piloting, which is good since the reagents expire within about a week of being opened. For our specific experimental conditions, GLP-1 was not particularly elevated, so we found that diluting plasma samples 2X beforehand brought us close to the bottom of the standard curve, but were still within range. Anyone using this kit should probably perform a quick pilot with diluted and non-diluted samples as well, but the standard curve is robust enough and covers a large enough range that I think it will work well for all study conditions. Finally, I found the instructions easy to follow as I am an experienced ELISA user, but an undergraduate in our lab was a little confused and required me to re-order some of the instructions. So, experiences graduate students and post-docs won't have any issues, but new users should probably be walked through the instructions before they thaw any reagents. Overall, I was very happy and will purchase this again for future studies!
