RayBio^® Glycobiology Arrays are a versatile and innovative series of products designed to detect glycosylation. Glycosylation plays an important role in a wide variety of disease processes including tumor cell metastasis, inflammation and bacterial pathogenesis. Glycocalyx, literally meaning ‘sugar coat’, is an extracellular polymeric coating surrounding many prokaryotic and eukaryotic cells consisting of glycoproteins, glycolipids, proteoglycans and glycosaminoglycans. The constituents of the glycocalyx play an important role in the process of cell signaling, virus transfection, and immunity. However, detection tools for the research of glycobiology are currently in very limited supply.

Taking advantage of advancements in microarray technology developed for antibody arrays, RayBiotech developed 3 different formats of glycobiology arrays to help researchers detect glycans and/or glycan-protein interactions. All 3 formats feature glass slide supports with fluorescent detection.

Glycan Arrays

We have developed the first commercially available glycan arrays for screening protein-carbohydrate interactions. The glycan arrays help researchers:

• Identify the glycans binding partners in biological samples.
• Identify whether target proteins are carbohydrate binding proteins.
• Probe binding of viruses and whole cells to glycans.
• Profile the substrate specificity of enzymes (glycosyltransferases, glycosidases, etc.).
• Profile the inflammatory immune response.

Glycome Arrays

The RayBio^® Glycome Arrays allow researchers to efficiently obtain glycosylation profiles in their samples. Capture antibodies are printed onto glass slides and the glycans on these capture antibodies are removed. The glass slide arrays come pre-blocked and are ready to be incubated with samples. After incubation with samples and washing to remove unbound proteins, five unique biotin-labeled lectins are incubated with the array to allow binding to the glycans from captured target proteins. Streptavidin-conjugated fluorescent dye (Cy3 equivalent) is then added to the array. Finally, the glass slide is dried and laser fluorescence scanning is used to visualize the signals. These signals are then compared to the array map to identify glycosylated proteins present in the samples.

Lectin Arrays

Lectins are glycan-binding proteins which have been purified from trees, beans and some fruits. They are highly specific for a given glycan based on their sequence and the different sugar unit structures the glycan contains. The RayBiotech lectin arrays provide a powerful new tool for glycosylation determination, drug discovery and biomarker development; all with limited samples volumes required.
