Introduction
The RayBio® Plasma Membrane, Cytosol and Nuclear Protein Extraction Kit provides a rapid and efficient method for enriching native plasma membrane, cytosolic, and nuclear protein fractions from cultured cells or tissue samples. The kit uses a specially formulated reagent system designed to reduce cross-contamination between fractions while maintaining high protein recovery. The enriched protein fractions are suitable for a wide range of downstream applications, including SDS-PAGE, Western blotting, ELISA, immunoprecipitation, membrane protein structural analysis, 2-D gel electrophoresis, enzyme activity assays, and other protein-based analyses. The procedure is simple to perform and can be completed in as little as 2 hours for up to 20 samples.

Additional Materials Required
- Pipette tips (barrier tips recommended) and microcentrifuge tubes
- PBS
- Microcentrifuge
- Water bath
- Vortex mixer
- Tissue homogenizer if using tissue samples (Dounce tissue grinder recommended)
- Refrigerated centrifuge with swing out rotor and refrigerated microcentrifuge
Assay Procedure Summary
- Prepare cultured cell or tissue samples as instructed.
- Prepare all reagents and keep samples and buffers on ice unless otherwise specified.
- Sequentially isolate the nuclear, cytosolic, and plasma membrane fractions.
- Store the enriched fractions at –80°C until use.
Storage/Stability
The kit can be stored at 4°C for a period of 6 months prior to opening. After Protease Inhibitor Cocktail has been reconstituted, store at -20°C or -80°C.
